ELISA Methods Based on Monoclonal Antibodies for the Serological Diagnosis of Lumpy Skin Disease

Lumpy skin disease (LSD) is a notifiable, transboundary cattle that spreads rapidly and has relevant economic impact. The etiological agent the LSD virus (LSDV), genus Capripoxvirus, family Poxviridae. To date, LSDV widely present in Africa, Asia, transcontinental regions like Russia, Turkey, Middle East, thus representing continuous threat to free neighbours. Appropriate serosurveillance programs can complement control, inform about its spread, enable assessment of vaccination campaigns. Since reliable practical diagnostic tools could improve serological surveillance, this study aimed produce characterize monoclonal antibodies (MAbs) allowed us develop ELISA tests for detection LSD. Four MAbs recognizing 35 kDa viral protein were selected used optimize competitive indirect ELISAs. Both assays detected seroconversion within 14 days postinfection (dpi) 18 experimentally infected with sequentially sampled up 4 weeks. two novel ELISAs also raised by other capripoxviruses: as observed cattle, both revealed dpi all nine sheep sheeppox (SPPV), while eight goats goatpox (GTPV) identified seropositivity earlier more animals compared ELISA. Overall, sensitivity performance developed resulted comparatively superior those neutralization test commercial Id.Vet Testing 200 negative sera from each species recorded single false-positive ELISA, which gave specificity 99.5%, whereas was 100% irrespective tested. results concluding new correctly detect anti-LSDV be recognize SPPV GTPV.
